What is the difference between Gram-positive and Gram-negative bacteria? Gram-positive bacteria show blue or purple after gram-staining in a laboratory test. They have thick cell walls. Gram-negative bacteria show pink or red on staining and have thin walls.
Gram positive bacteria possess a thick (20–80 nm) cell wall as outer shell of the cell. In contrast Gram negative bacteria have a relatively thin (<10 nm) layer of cell wall, but harbour an additional outer membrane with several pores and appendices.
Actinobacteria is the taxonomic name of the class of high G+C gram-positive bacteria. This class includes the genera Actinomyces, Arthrobacter, Corynebacterium, Frankia, Gardnerella, Micrococcus, Mycobacterium, Nocardia, Propionibacterium, Rhodococcus, and Streptomyces.

Gram-negative bacteria, on the other hand, have a thin peptidoglycan layer that is covered by multiple thin layers of membrane that can better-resist antibiotics.
Escherichia coli (E. coli) is a Gram-negative, rod-shaped, facultative anaerobic bacterium.
Gram-negative bacteria are more harmful than gram-positive bacteria. This is because gram-negative bacteria have a hard, protective outer shell, which makes them more resistant to antibiotics and hence harder to kill.

The organisms are identified based on color and shape. Gram-positive organisms are either purple or blue in color, while gram-negative organisms are either pink or red in color. Bacilli are rod-shaped, while cocci are spherical.
A mixture of hydrogen peroxide and ascorbic acid has been found to generate an antibacterial mechanism which is active against gram-negative bacteria. It results in bacterial death and renders the organism sensitive to lysis by lysozyme.
